Christian@mabtech.com Posted September 11, 2014 Report Share Posted September 11, 2014 Dear Raimond, When analyzing spots in ELISpot where the plates have been developed under the same experimental conditions, the fundamentals of are of course there: a large spot correspond to a higher level of secretion compared to a small spot. However, determining anything beyond this basic point of reality is difficult to do in a quantitative and reliable manner. You see, our experience is that the dynamic range in this aspect is poor. ELISpot is an excellent tool for determining the number of cytokine secreting cells but less appropriate for making judgments on how much cytokine is actually released by the activated cell. It is often so that quite marginal differences in spot-size in reality correspond to quite large differences in the amount of cytokine secreted.
